摘  要:目的探讨BTB-CNC异体同源体1(Bach1)和γ-谷氨酰半胱氨酸合成酶(γ-GCS)在支气管哮喘豚鼠肺组织和肺泡灌洗液(BLAF)炎症细胞中的表达。方法 38只健康雄性豚鼠按随机数字表法随机分为对照组、哮喘组和地塞米松治疗组,卵蛋白致敏法复制哮喘模型,检测3组豚鼠肺组织匀浆中丙二醛浓度。RT-PCR和Western blot法分别检测肺组织中Bach1、γ-GCS mRNA和蛋白;原位杂交法和免疫细胞化学法检测BLAF炎症细胞中Bach1、γ-GCS mRNA和蛋白。结果肺组织中丙二醛浓度哮喘组高于对照组和地塞米松治疗组。肺组织和BALF炎症细胞中γ-GCS mRNA和蛋白表达哮喘组低于对照组和地塞米松治疗组;Bach1 mRNA三组间差异无显著性;Bach1蛋白细胞核内表达哮喘组高于对照组和地塞米松治疗组。肺组织和BALF炎症细胞中γ-GCSmRNA和蛋白表达呈正相关;肺组织和BALF中γ-GCS mRNA表达与Bach1核内表达呈负相关。结论肺组织和BALF炎症细胞中γ-GCS、Bach1表达结果一致;支气管哮喘急性期豚鼠肺组织和BALF炎症细胞中γ-GCS表达较低,而细胞核内Bach1蛋白表达增加,地塞米松增加哮喘豚鼠的抗氧化能力可能与调控Bach1核转位而增加γ-GCS表达有关。Objective To investigate the expression of BTB and CNC homology 1(Bach1) andγ-Glutamylcysteine synthetase(γ-GCS) in lung tissues and inflammatory cells from guinea pigs with bronchial asthma. Methods 38 healthy adult male guinea pigs were randomly divided into control group,asthmatic group and dexamethasone group.Asthmatic model was established through ovalbumin challenge method.The malondialdehyde concentration of the lung tissue homogena was detected in three groups.The mRNA and protein expression of Bach1 and γ-GCS in lung tissues were measured by RT-PCR and Western blot,respectively.The methods of in situ hybridization and immunohistochemistry were used for detection of the mRNA and protein expression of Bach1 and γ-GCS in inflammatory cells in BALF. Results The malondialdehyde concentration(nmol/mg)of lung tissue from guinea pigs in group B was significantly higher than that in group A and group C.The expression ofγ-GCS mRNA in group B was significantly lower than that in group A and group C in lng tissues and inflammatory cells in BALF.The mRNA ecpression Bach1 had no significant difference among the three groups.But the expression of Bach1 in cell nucleus in group B was higher than that in group A and group C.There was a positive correlation between the expression of γ-GCS mRNA and protein,but a negtive correlation between the expression of γ-GCS mRNA and the expression of Bach1 in cell nuclus in lung tissues and inflammatory cells of BALF. Conclusions The expression of Bach1 and γ-GCS in lung tissues and inflammatory cells in BALF is consistent.The expression of γ-GCS is low in lng tissues and inflammatory cells in BALF but expression of Bach1 protein in cell nucleus is increaed from acute stage of bronchial asthma.The capability of anti-oxidation of guinea pigs being increased by dexamethasone may be due to regulating Nrf2/Bach1 nuclear translocation and increasing expression of γ-GCS.
