针刺对脑缺血再灌注模型大鼠脑组织基质金属蛋白酶、细胞间黏附分子表达的影响

摘　　要：目的探讨针刺对脑缺血再灌注模型大鼠脑损伤区组织基质金属蛋白酶(MMP-9)和细胞间黏附分子-1(ICAM-1)表达的影响。方法以大脑中动脉栓塞法制备脑缺血再灌注模型,采用随机区组设计将大鼠分为电针、针刺、药物、针药结合、模型、假手术等6个大组,其中电针、模型、假手术组又分12、24、48、72、96和144 h 6个时间点以,另设空白对照组7只。HE染色检测损伤区组织形态学变化;免疫组化法检测MMP-9和ICAM-1积分吸光度值。结果 HE染色显示,梗死核心区病理损伤高峰时间出现在48 h;72 h后间质水肿减轻,大量小胶质细胞和胶原纤维增生,出现大量新生血管。免疫组化结果显示,MMP-9和ICAM-1在血管内皮细胞和炎性细胞及少数神经元广泛表达,缺血区明显高于自身对照区;组内比较显示表达呈48 h和144 h双峰形;组间比较显示,在48、72和96 h,电针组与其他实验组结果有显著差异。结论电针可能通过上调损伤-修复网络功能,促进梗死区神经功能的恢复。Objective To study the cerebral expression of MMP-9 and ICAM-1 in rats treated with acupuncture after cerebral ischemic /reperfusion.Methods SD rats were randomly divided into six groups,electro-acupuncture at ＂Zusanli＂ and ＂Bai hui ＂ group,＂Baihui＂ group,Nimodipime group,joint treatment group of acupuncture and medicine,model,sham group and another 7 as control group.Electro-acupuseture,model and sham group contain 12,24,48,72,96 and 144 h six time points.The pathological lesion was detected by HE staining.The expression of MMP-9 and ICAM-1 in core infarct area was examined by immunohistochemistry staining.Results That the pathological lesion reached the peak at 48th hour in the infarct core area;cerebral interstitial oedema was not alleviated until 72 hrs.Also many glial cells and collagen fibers proliferate,and new blood capillaries appear.MMP-9 and ICAM-1 was widely expressed in vascular endothelial and inflammatory cells together with some neurons by immunohistochemistry staining.The positive cells in the ipsilateral region is more than in contralateral region;Com-parison within groups shows MMP-9 and ICAM-1 expression at 48th and 144th hours with a double-peak profile,while the expression in electro-acupuncture group at 48,72 and 96 h time points has significant difference compared with that in model group.Conclusion Acupuncture may promote neurological function recovery through up-regulating injury mediation network.
