摘  要:目的探讨间断性和持续性阻断肝血流对线粒体功能的影响及其作用机理。方法实验大鼠分为持续阻断组、间断阻断组和假处理组,检测其肝线粒体功能、肝组织内钙和脂质过氧化物(Lipidperoxide,LPO)含量,以及超氧化物歧化酶(superoxidedismutase,SOD)活性。结果与间断性阻断肝血流相比,持续性缺血再灌注后大鼠肝线粒体呼吸控制比率(respiratorycontrolratio,RCR)、磷氧比值(P/O)、氧化磷酸化效率(oxidativephosphorylationrate,OPR)和SOD活性均显著下降,其值分别为304±017对209±015(P<001)、151±009对122±010(P<005)、242±23对134±23(P<001)及5734±385U·mgpr-1对4229±336U·mgpr-1(P<005);而肝组织内钙与LPO值均显著升高,分别为372±24μg/g对485±29μg/g(P<005)及520±017nmol·mgpr-1对587±017nmol·mgpr-1(P<005)。而在间断性血流阻断组与假处理组之?Objective To evaluate the effect of discontinuous and continuous interruption of blood supply to the liver on mitochondrial function in rat.Methods Rats were divided into three groups:discontinuous interruption,continuous interruption and sham operation.Liver mitochondrial function,contents of calcium and lipid peroxide (LPO),and superoxide dismutase (SOD) activity of the rat were measured.Results Compared with discontinuous blood supply interruption,respiratory control ratio (RCR),P/0 ratio, oxidative phosphorylation rate (OPR) and SOD activity significantly declined and contents of calcium and LPO increased after continuous ischemia reperfusion.They were 3 04±0 17 vs 2 09±0 15 ( P <0 01),1 51±0 09 vs 1 22±0 10 ( P <0 05),242±23 vs 134±23 ( P <0 01),5734±385 U·mgpr -1 vs 4229±336 U·mgpr -1 ( P <0 05),37 2±2 4 μg/g vs 48 5±2 9 μg/g ( P <0 05) and 5 20±0 17 nmol·mgpr -1 vs 5 87±0 17 nmol·mgpr -1 ( P <0 05),respectively.All parameters in discontinuous interruption group have changed insignificantly as compared with sham operative rats.Conclusions The results suggest that discontinuous ischemia reperfusion of liver is less hazardous to mitochondrial function because the calcium equilibrium and activity of SOD remained comparatively normal.
