组胺H_3受体激动剂IMETIT对豚鼠变应性鼻炎作用的初步探讨

摘　　要：目的:探索组胺H3受体激动剂(H3R激动剂)IMETIT对豚鼠变应性鼻炎(AR)模型的治疗作用和对P物质(SP)及其受体(SP-R)mRNA表达的影响。方法:以OVA为致敏原,用豚鼠AR模型,观察症状及鼻黏膜病理状况以研究IMETIT对豚鼠AR的治疗作用;免疫组织化学检测鼻中隔黏膜中SP含量,RT-PCR测定鼻中隔黏膜中SP-RmRNA表达以研究IMETIT对SP/SP-R分泌及表达的影响。结果:组胺H3R激动剂IMETIT能有效改善豚鼠AR喷嚏(P<0.01)、鼻痒(P<0.01)、鼻塞(P<0.05),减轻鼻黏膜病理性改变,调低SP/SP-RmRNA分泌及表达(P<0.05或P<0.01)。结论:组胺H3R激动剂IMETIT能有效缓解豚鼠AR的症状,其作用机制与调低SP/SP-R分泌及表达有关。Objective：To discuss the treatment of H3R agonist,IMETIT,on the allergic rhinitis（ AR） ,and the influence to mRNA of Substance P（ SP） and Substance P Receptor （ SP-R） in AR model of guinea gigs. Method：The severity of AR was assessed by allergic symptoms（ sneezing,nasal rubbing and nose blocking）. The changes in the nasal mucosa were studied by pathological methods. The expression of SP positive cell was detected by immunohistochemistry,and the expression of SP-R mRNA was detected by reverse transcriptive polymerase chain reaction（ RT-PCR）. Result：Histamine H3R agonists,IMETIT can effectively improve the AR symptoms,sneezing,nasal itching,nasal congestion,reduce the pathological changes in the nasal mucosa,cut down the SP secretion and SP-R mRNA expression. Conclusion：Histamine H3R agonist,IMETIT can effectively relieve the symptoms of AR in guinea pigs,which is related to reducing SP secretion and SP-R mRNA expression.
