摘 要:丙酮酸激酶(Pyruvate kinase,PK)为催化糖酵解途径中最后一步反应关键酶,是植物能量释放利用的关键步骤。本研究采用生物信息学方法对水稻基因组进行扫描,获得10个具有PK极保守区域(PK-PK-C)的基因。进行氨基酸序列比对结果表明该家族所有基因的氨基酸一致性为38.4%,系统进化树分析结果也暗示该家族存在4对旁系同源基因;MEME程序分析结果表明该家族都具有完全保守的基序,这些基序排列次序基本相同。对预测定位在线粒体的4个PK基因进行半定量RT-PCR分析,结果表明,Os04g0677500基因在各器官的表达量都很高,呈组成性表达;而Os10g0571200在各器官基本不表达;Os01g0276700和Os03g0672300表达在各器官中存在差异性。4个mtOsPKs表达情况的多样性可能是水稻在进化中适应不同生存环境而产生,使线粒体作为能量加工厂的正常运转提供保证。Pyruvate kinase (PK)is a key enzyme involved in glycolysis which is the final step in the glycolytic pathway.Glycolytic pathway is the key step involved in yield and utilization of energy of plant.Sequence alignment indicated that the amino acids identity encoded by PK genes was 38.4%.Phylogenetic analysis suggested the existence of four pairs of paralogous genes in the family.MEME program analysis showed that the family had a highly conserved motif.Semi-quantitative RT-PCR of the four PK genes predicted to locate in mitochondria showed that the expression of Os04g0677500 is high in various organs, indicating that the constitutive expression of the gene Os04g0677500, whereas Os01g0276700 and Os03g0672300 was differentially expressed in various organs.Almost no expression of Os10g0571200 is detected in various organs.The diversity of expression pattern of the four mtOsPKs implied that rice had evolved PK genes which guarantee the normal operation of mitochondria as an energy processing plantto adapt to different living environment,
