高效液相色谱法检测大鼠血浆中盐酸环苯扎林浓度的方法研究

摘  要:目的:建立血浆中盐酸环苯扎林含量的测定方法。方法:用高效液相色谱法(HPLC),色谱柱为HypersilBDS-C18(5μm,4.6mm×250mm),乙腈-0.085%磷酸水溶液(三乙胺调pH为6.5)65:35为流动相;流速为1.0mL·min-1;检测波长290nm。结果:盐酸环苯扎林在12.06~241.2ng·mL-1范围为内与峰面积线性关系良好(r=0.9998);对照品溶液48小时内稳定,RSD为1.1%;低、中、高3种血浆浓度的平均回收率为(94.45±5.21)%、(98.64±2.74)%和(99.62±1.36)%。结论:该方法简便,精密度及重现性均较好,分离效果好,该法适应于盐酸环苯扎林血浆样品的快速、大量分析。Objective:To study the determination method of cyclobenzaprine hydrochloride in rat plasma by HPLC.Methods:The separation is performed by Hypersil BDS-C18(5 μm,4.6 mm×250 mm)with the mobile phase of acetonitrile-0.085% phosphoric acid aqueous solution adjusted pH 6.5 by triethylamine (65:35V/V).While the detection wavelength was 290nm for the determination of cyclobenzaprine hydrochloride.Results:Cyclobenzaprine hydrochloride and other peaks were separated well and the peak shape was presented well.There was the good linearity within the range between 12.06 ng·mL-1~241.2 ng·mL-1 of cyclobenzaprine hydrochloride (r=0.9998).The reference substance solution presented fair stabilization within 48 h and RSD was 1.1%.The average recovery rates for low,middle and high solutions were (94.45±5.21)%,(98.64±2.74)% and (99.62±1.36)respectively.Conclusion:The method is sensitive,accurate with good reproducibility.And it is suit for the massive and rapid determination of cyclobenzaprine hydrochloride plasma samples.
