隐球菌性脑膜炎患者外周血中TLR4的表达以及血清中TNF-α，IL-10的水平研究

摘　　要：目的探讨Toll样受体4（TLR4）在隐球菌性脑膜炎患者外周血中的表达，以及研究血清中TNF—α，IL-10的变化情况。方法收集25例隐球菌性脑膜炎患者和25例健康体检者全血，采用荧光定量PCR（FQ—PCR）检测TLR2，TLR4的mRNA表达。ELISA法检测血清中TNF—α，IL-10的表达。结果隐球菌性脑膜炎患：者TLR4 mRNA的表达显著低于对照组（0．35±0．18）（P〈0．05），TLR2均无显著变化（0．87±0．23）（P〉0．05）。血清中的IL-10显著升高（4．25±1．30）pg／ml（P〈0．05），TNF—α的变化不显著（1．49±0．87）pg／ml（P〉0．05）。结论隐球菌性脑膜炎患者TLR4 mRNA表达显著下降，可能在隐球菌性脑膜炎的发病过程中起一定的作用。Objective To investigate expression of TLR4 in peripheral blood from patients with Cryptococcal Meningitis, and detect the levels of IL-10 and TNF-α in serum. Methods Peripheral blood of 25 patients with Cryptocoecal Meningitis and 25 health donors were collected ,mRNA expressions of TLR2 and TLR4 were detected with FQ-PCR. Levels of TNF- α and IL-10 in the serum were examined with ELISA. Results Compared with control,mRNA level of TLR4 gene in patients with Cryptoeoccal Meningitis markly increased（0.35±0. 18） （P〈0. 05）,but level of TNF-α had not significant difference（1.49±0. 87）pg/ml （P〉0. 05）. Conclusion Change of mRNA expressions of TLR4 gene in patients with Cryptococcal Meningitis may play the important role in process of Cryptococcal Meningitis.
