摘  要:目的探讨氧化低密度脂蛋白(ox-LDL)诱导病理免疫的可能机制及雷公藤多甙(TG)的干预作用。方法采集6例健康志愿者外周静脉血,密度梯度离心法分离单个核细胞,然后分为5组进行体外孵育:ox-LDL组(ox-LDL终浓度1mg/L)、TG高剂量组(TG和ox-LDL终浓度分别为1.5mg/L和1mg/L)、TG中剂量组(TG和ox-LDL终浓度均为1mg/L)、TG低剂量组(TG和ox-LDL终浓度分别为0.5mg/L和1mg/L)、对照组(不加ox-LDL和TG)。采用流式细胞术检测各种淋巴细胞亚群比例,ELISA法检测细胞培养上清液中人氧化低密度脂蛋白抗体(OLAb)浓度。结果对照组T细胞、Th细胞、Tc细胞和Treg细胞的活化率分别为2.04%±0.54%、1.12%±0.18%、1.58%±0.77%和13.04%±7.84%。与对照组比较,ox-LDL组T细胞活化率(4.37%±1.10%)、Th细胞活化率(2.61%±0.88%)和Tc细胞活化率(3.64%±1.96%)均明显升高(P<0.05),Treg细胞活化率(17.29%±9.88%)差异无统计学意义(P>0.05)。与对照组比较,ox-LDL组Tc细胞活化率升高幅度(2.07%±1.93%)与Th细胞活化率升高幅度(1.49%±0.74%)差异无统计学意义(P>0.05)。TG中剂量组T细胞、Th细胞和Tc细胞的活化率分别为1.71%±0.48%、1.38%±0.22%和1.56%±0.28%,均显著低于ox-LDL组(P<0.05),而与对照组比较差异无统计学意义(P>0.05),Treg细胞活化率(20.01%±11.41%)显著高于对照组(P<0.05),与ox-LDL组比较差异无统计学意义(P>0.05)。与ox-LDL组比较,TG中剂量组Tc细胞活化率降低幅度(2.09%±1.83%)与Th细胞活化率降低幅度(1.23%±0.95%)比较差异无统计学意义(P>0.05)。ox-LDL组OLAb浓度(443.82±132.46μg/L)显著高于对照组(183.86±88.49μg/L,P<0.05)。TG高、中、低剂量组OLAb浓度(分别为183.89±101.64、222.51±62.71、249.23±84.95μg/L)与对照组比较差异无统计学意义,且三个剂量组间差异也无统计学意义(P>0.05),但与ox-LDL组比较,三个剂量组OLAb浓度均明显降低(P<0.05)。结论人淋巴细胞受ox-LDL刺激后能产生自身免疫样反应,TG�Objective To investigate the mechanism of immunodysfunction induced by oxidized low density lipoprotein (ox-LDL),and to observe intervention effect of tripterygium glycosides (TG) on the condition.Methods Venous blood samples of 6 healthy volunteers were collected,and mononuclear cells were isolated by density gradient centrifugation,and then cultured and grouped as:ox-LDL group,high dose tripterygium (HDT) group,moderate dose tripterygium (MDT) group,low dose tripterygium (LDT) group and control group.Quantity and percentage of all kinds of lymphocyte were counted by flow cytometry,and the concentration of ox-LDL antibody (OLAb) in supernatant was assayed by ELISA.Results Activation ratio of T cell,Th cell,Tc cell and Treg cell of control group was 2.04%±0.54%,1.12%±0.18%,1.58%±0.77% and 13.04%±7.84%,respectively.Compared with that of control group,the activation ratios of T cell (4.37%±1.10%),Th cell (2.61%±0.88%) and Tc cell (3.64%±1.96%) of ox-LDL group were all increased significantly (P〈0.05).No significant difference on Treg cell activation ratio was found between ox-LDL group (17.29%±9.88%) and that in control group (P〉0.05).Compared with control group,no significant difference was found in the increased extent of activation ratio between Tc cell (2.07%±1.93%) and Th cell (1.49%±0.74%) in ox-LDL group (P〉0.05).Activation ratios of T cell,Th cell and Tc cell (1.71%±0.48%,1.38%±0.22% and 1.56%±0.28%,respectively) in MDT group were less than that in ox-LDL group (P〈0.05),but no significant difference was found between MDT group and control group (P〉0.05).Treg cell activation ratio in MDT group (20.01%±11.41%) was significantly higher than that in control group (P〈0.05),but no significant difference was found between MDT group and ox-LDL group (P〉0.05).No significant difference in activation ratio in decreased extent was found between Tc cell (2.09%±1.83%) and Th cell (1.23%±0.95%) in MD
