摘 要:目的观察流式细胞术(FCM)检测网织红细胞(RET)与传统方法的优势及其检测信息与临床的联系。方法以噻唑橙作为RNA的荧光染料标记RET,采用FCM测定RET和网织血小板(RP)的百分数、RNA残留荧光强度;传统RET测定以煌焦油蓝染色、显微镜计数观察。结果 FCM法与传统方法比较,其重复性和准确性差异无统计学意义(P>0.05),但FCM的变异系数小于传统方法 .300例研究对象间各贫血组RET%和平均荧光强度与健康对照组比较差异均有统计学意义(P<0.05)。100例血小板减少患者其RP%和平均荧光强度与健康对照组比较差异有统计学意义(P<0.05)。结论 FCM测定RET与传统方法相比具有准确性高、重复性好等特点,在检测RET%时能同时检测到RET的平均荧光强度和RP,平均荧光强度可反映RET和RP的成熟情况。Objective To observe the superiority of reticulocyte detection by the flow cytometry (FCM) to tradition method and the connection of its detected information with clinic.Methods Reticulocyte was determined by the flow cytometry with thiazole orange as marker,the percentage of reticulocyte,reticulated platelet and intensity of RNA remaind fluorencence were detected tradition method used microscope.Results The repeatability and accuracy had no significant differences between the methods of flow cytometry and tradition,but in the flow cytometry coefficient of variation was lower than that in the tradition method.In 300 objects of the anaemia group,fluorencence intensity and percentage of reticulocyte had significant difference compared with health group (P〈0.05).In 100 cases of thrombocytopeina,fluorencence intensity and percentage of reticulated platelet had significant difference compared with the health group (P〈0.05).Conclusion Compared with the tradition method,the flow cytometry has the advantages of good repeatability and high accuracy,which can simultaneously determine the average fluorencence intensity of reticulocyte and reticulated platelet,the average flnorencence intensity of reticulocyte could reflect their maturity.
