摘 要:目的:探讨氧自由基在肝缺血再灌注损伤(HIRI)中的作用及川芎嗪的防护作用。方法:应用HIRI家兔及肝癌手术患者,观察超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX),黄嘌呤氧化酶(XO)、谷丙转氨酶(ALT)活性、丙二醛(MDA)含量、肝细胞形态学变化及川芎嗪对上述指标的影响。结果:HIRI时SOD、GSH-PX活性显著下降(P<0.05和P<0.01),XO、ALT活性及MDA含量明显升高(P<0.05和P<0.01),肝细胞形态学发生异常变化;使用川芎嗪后,上述指标的异常变化均明显减轻,其差异有显著和非常显著(P<0.05和P<0.01)。结论:氧自由基是HIRI的重要发病学因素,而川芎嗪通过清除氧自由基可减轻HIRI。AIM:To explore the role of oxygen free radicals (OFR) in hepatic ischemia-reperfusion injury (HIRI) of rabbits and humans and the protective effect of ligustrazin (LGT). METHODS: Changes of several parameters and effects of LGT on it were observed during HIRI in 20 rabbits and 18 patients who suffered from cancer of the liver and were scheduled for elective hepatic surgery, parameters of which included superoxide dismutase (SOD) glutathione peroxidase (GSH-PX) xanthine oxidase (XO) alanine aminotransferase (ALT) and malondialdehyde (MDA) as well as morphological changes of hepatocytes.RESULTS: SOD and GSH-PX activities decreased significantly ( P <0.05) and very significantly ( P <0.01), XO and ALT activities as well as MDA concentration increased remarkably ( P <0.05 and P <0.01), and abnormal morphological changes of hepatocytes were obvious during HIRI; with treatment of LGT, the abnormal changes of all above parameters were diminished remarkably ( P <0.05 and P <0.01). CONCLUSION: It is indicated that OFR may be one of the primary causes for HIRI, LGT can attenuate HIRI by eliminating OFR.
