摘 要:目的建立茄根中绿原酸含量的反相高效液相色谱(RP-HPLC)测定法。方法色谱柱为YMC-Pack ODS-AA(150 mm×4.6 mm,5μm),以乙腈-甲酸水的梯度流动相洗脱,检测波长323 nm,流速1 ml.m in^-1,柱温30℃。结果在线性范围内(0.005-0.04 mg.m l^-1)绿原酸对照品的标准曲线呈良好的线性关系(r=0.999 8),茄根中绿原酸的含量为0.108 mg.g^-1,RSD为2.4%;平均回收率为98.51%,RSD为2.7%。结论可通过RP-HPLC测定茄根中绿原酸含量,为茄根的进一步开发利用提供科学依据。Objective To establish a method for determination of chlorogenic acid content in the dried root of Solamum melongena L..Methods Chromatographic condition included a YMC-Pack ODS-AA(150 mm×4.6 mm,5 μm),a gradient solvent system of acetonitrile and water was applied from 10% to 60% of acetonitrile within 30 minutes with the detection wavelength at 323 nm.The flow rate was 1.00 ml·min^-1 and column temperaturewas set at 30 ℃.Results The linear relationship of chlorogenic acid was determined within the range from 0.005 to 0.04 mg · ml^-1(r = 0.999 8).Using the method above,the content of the chlorogenic acid was determined as 0.108 mg · g^-1,RSD was 2.4%,with the recoveries as 98.51% with RSD was 2.7%.Conclusion A convenient and reliable method was established to determine the content of chlorogenic acid in the dried root of Solamum melongena L..
