摘  要:目的建立一种快速、准确测定人血清中霉酚酸浓度的超高效液相色谱-质谱(UPLC-MS)测定方法。方法在血清中加入内标吲哚美辛,采用超高效液相色谱联合质谱的方法对血清中的霉酚酸进行分离测定。结果霉酚酸在0.05~100.00μg/mL的浓度范围内呈良好线性,r=0.9980,最低检测线为0.05μg/mL,方法回收率大于91.2%,日内、日间变异系数均小于5%。结论该方法准确、快速、灵敏,可用于霉酚酸的治疗药物监测、人体内药代动力学及生物等效性研究。Objective To establish a rapid and accurate method using UPLC-MS for quantification of mycophenolic acid in human serum.Methods Indometacin was added to human serum sample and mycophenolic acid in human serum was determined by UPLC-MS.Results The calibration curve was linear(r=0.9980)among the concentration range of 0.05~100.00 μg/mL and the limit of the quantitative detection was 0.05 μg/mL.The recovery of the method was higher than 91.2%.The coefficients of variation for inter-day and intra-day were both lower than 5%.Conclusion As an accurate,rapid and sensitive method,this assay can be used in the supervision of mycophenolic acid drug and the study of pharmacokinetics and bioavailability.
