HCN4基因在风湿性心脏病二尖瓣狭窄伴心房颤动心房组织中蛋白表达的研究

摘　　要：目的:通过检测风湿性心脏病二尖瓣狭窄伴心房颤动(房颤)患者心房肌细胞内HCN4蛋白表达量,探讨风湿性心脏病二尖瓣狭窄伴房颤的发生机制。方法:选取风湿性心脏病二尖瓣狭窄伴房颤患者30例为实验组,同期取风湿性心脏病二尖瓣狭窄非房颤19例为对照组,通过Western blot检测心房肌细胞内HCN4蛋白表达量。结果:实验组和对照组中HCN4基因在心房肌组织中的蛋白表达分别是0.29±0.03和0.13±0.03,两组比较,差异有统计学意义(P<0.01)。结论:风湿性心脏病二尖瓣狭窄伴房颤患者心房肌组织中确实存在HCN4基因的表达上调,提示HCN4基因的过度转录表达可能参与了调控风湿性心脏病房颤的发生过程。Objective：Through the quantitative analysis of HCN4 gene expression in muscles of right atrium in atrial fibrillation patients with mitral valve stenosis of rheumatic heart disease, we aim to explore the mechanism of atrial fibrillation in mitral valve stenosis of rheumatic heart disease. Methods： Western blot was used to study HCN4 gene expression in two groups of rheumatic heart disease patients with mitral valve stenosis, one group with atrial fibrillation （exprerimental group, n =30） and another group without atrial fibrillation （control group, n = 19）. Results：The relative expression value of HCN4 gene in experimental group and control group was 0.29±0. 03 and 0.13±0.03（ P 〈0.01） ;respectively. Conclusion：The results confirmed that the HCN4 gene expression rises in atrial fibrillation patients with mitral valve steno- sis of rheumatic heart disease. It also indicates that the overexpression of HCN4 gene might participate in the progress of atrial fibrillation development in mitral stenosis of rheumatic heart disease.
