摘  要:目的研究藻蓝蛋白对2型糖尿病大鼠心肌细胞结构及诱导型一氧化氮合酶(iNOS)表达的影响,探讨可能的作用机制。方法健康雄性Wistar大鼠32只,适应性喂养1周后,随机选择8只为正常组(A组),其余24只应用高脂高糖饲料喂养4周,联合30 mg/kg链脲佐菌素(STZ)腹腔注射方法建立2型糖尿病大鼠模型,造模成功后,将其随机分为模型组(B组)、藻蓝蛋白治疗组(C组)及二甲双胍治疗组(D组),各8只,C、D组分别用藻蓝蛋白及二甲双胍灌胃治疗10 d,生化方法测定各组大鼠空腹血糖,组织病理学技术观察各组大鼠心肌细胞的形态结构,免疫组化技术检测各组大鼠心肌细胞内iNOS的表达。结果造模成功后,B、C、D组空腹血糖与A组相比明显升高(F=17.54,q=7.73~8.72,P〈0.01);治疗后C、D组大鼠空腹血糖较B组明显降低,差异有显著性(F=21.79,q=4.16、7.03,P〈0.01);C、D组大鼠心肌细胞的病理形态及结构较B组有较大改善;C、D组大鼠心肌细胞内iNOS阳性细胞数与B组相比明显减少(F=180.30,q=18.88、24.40,P〈0.01),而C组与D组大鼠心肌细胞iNOS阳性细胞数比较差异无显著性(P〉0.05)。结论藻蓝蛋白可降低糖尿病大鼠空腹血糖水平,改善心肌细胞损伤,并抑制糖尿病大鼠心肌细胞iNOS表达,其作用可能通过抗氧化机制,保护糖尿病状态下的心肌细胞。Objective To investigate the effect of phycocyanin on the cellular structure and the expression of inducible nitic oxide synthase(iNOS) in myocardial cells in rats with type 2 diabetes mellitus. Methods Thirty-two healthy male Wistar rats were adaptively fed for one week and eight of them were then randomly selected to serve as normal control group(A),the rest were fed with high-fat and high-glucose forage for four weeks by combination of intraperitoneal injection of(30 mg/kg) streptozotocin(STZ) to create a model of type 2 diabetes in rats.Upon completion of the models,the 24 rats were evenly randomized to diabetes-model group(B),phycocyanin group(C) and metformin group(D).Intragastric phycocyanin was offered to group C,and metformin to group D,respectively,for 10 days.The fasting blood glucose(FBG) was determined by biochemical assay,the morphosis of cardiac myocytes observed by histopathological technique,and the expression of iNOS within cardiac myocytes detected immunohistochemically. Results After creation of the model,FBG of groups B,C and D was significantly higher than that of group A(F=17.54;q=7.73,8.54,8.72;P0.01);Post-therapeutically,the FBG of groups C and D was obviously lower than that of group B(F=21.79;q=4.16,7.03;P0.01);Morphologically,the cardiac myocytes of rats in groups C and D much improved than those in group B.Compared with group B,the expression of iNOS in groups C and D was weaker(F=180.30;q=18.88,24.40;P0.01),but the difference of that between the groups C and D was not significant. Conclusion Phycocyanin can lower the FBG,improve the pathologic damage of cardiac myocytes,and inhibit the expression of iNOS in diabetic rats,the action in which might be through antioxidation to protect cardiac myocytes under the status of diabetes.
