摘 要:目的研究低功率He-Ne激光照射对Jurkat细胞CD59活化途径的影响。方法将前期培养的Jurkat细胞随机平均分为3组:A组细胞不做任何处理,B组细胞每日定时进行7 min的He-Ne激光照射,C组细胞每日定时进行10 min的He-Ne激光照射。1周后用CD59单克隆抗体作用Jurkat细胞,用MTT法检测Jurkat细胞增殖情况,激光共聚焦方法观察细胞内Ca2+浓度变化,流式细胞术检测Jurkat细胞CD25表达。结果B、C组Jurkat细胞增殖率、Ca2+浓度和CD25表达均高于A组(F=41.390~63.938,q=10.907~24.497,P〈0.01);B组和C组间比较差异无显著性(P〉0.05)。结论低功率He-Ne激光照射可以促进Jurkat细胞经由CD59单克隆抗体刺激后的活化,增强活化信号的传导。Objective To study the effect of low-power He-Ne laser irradiation on CD59 activate channel in Jurkat cells.Methods Prophase cultural Jurkat cells were evenly randomized to three groups: group A was served as normal control;group B,Jurkat cells were irradiated 7 min daily;group C,the cells were irradiated 10 min daily.One week later,CD59 monoclonal antibody was applied to act on Jurkat cells,which were detected by transformation test(MTT assay).Laser confocal technique was employed to observe the changes of calcium ion concentration(CIC) within the cells;flow cytometry to detect the expression of CD25 in Jurkat cells. Results The growth rate of Jurkat cells,CIC and expression of CD25 in groups B and C were all higher than that in group A(F=41.390-63.938,q=10.907-24.497,P0.01).As compared between groups B and C,the difference being not significant(P0.05). Conclusion Low-power He-Ne laser radiation enhances the activation of Jurkat cells that stimulated by CD59,strengthening the conduction of activation signals.
