摘  要:本试验建立了不同温度、不同时间热应激实验小鼠动物模型,采用Real-time PCR和Western-blot方法检测了小鼠睾丸组织热应激蛋白70(HSP70)在mRNA和蛋白质水平的表达规律。结果表明:25℃时,HSP70在mR-NA和蛋白质表达水平没有变化;在31℃、34℃和37℃时,随应激时间的延长,HSP70mRNA和蛋白质的表达均呈现先升高后下降的趋势;40℃时,HSP70mRNA和蛋白质始终持续高表达。结论:小鼠受热应激后,睾丸组织HSP70mRNA和蛋白质均随着温度和时间的增加呈规律性变化,本研究为探索HSP70对精液品质的影响奠定了基础。In this study,we established the mouse models under different temperatures,and detected the mRNA and protein levels of heat shock 70 kDa protein (HSP70) in testicular tissue using Real-time PCR and Western-blot. Results showed that at 25℃,both the mRNA and protein levels of HSP70 kept at normal level. For the treat-ment groups of 31℃,34℃ and 37℃,the expression of HSP70 increased with processing time at first and reached to the highest level at 5 to 7 days,then reduced to the normal level. In 40℃ group,stable higher expression of HSP70 was observed. The above results indicated that the mRNA and protein levels of HSP70 in mouse testis have certain regularity under heat stress which can be one of the basic foundations for the further study of the thermal stress on the semen quality of large animal.
