摘  要:观察COX-2选择性抑制剂塞来西布(Celecoxib)联合外照射对5种不同恶性肿瘤细胞杀灭作用的影响,探讨塞来西布对恶性肿瘤细胞放射增敏作用的普遍性,为临床使用塞来西布作为放射增敏剂提供理论依据。选择适当浓度的COX-2选择性抑制剂塞来西布(Celecoxib)进行放射增敏实验,设置单纯放射组(R)和放射+药物(D+R)组,采用集落形成法分别检测Celecoxib对脑胶质瘤细胞株SHG-44、767,肺腺癌细胞株A549,宫颈癌细胞株HeLa,鼻咽癌细胞株CNE的放射增敏效应,并绘制细胞存活曲线。集落形成试验结果及存活曲线的相关参数显示,Celecoxib在2个胶质瘤株(SHG-44细胞、767细胞)均显示出放射增敏效应,R组和R+D组相比较,反映放射敏感性指标的SF2、D0.01、、D0、Dq均出现不同程度的下调。COX-2抑制剂Celecoxib在体外实验中可以提高多种恶性肿瘤细胞的放射敏感性。Celecoxib有可能成为治疗恶性肿瘤的放射增敏剂。COX-2 plays an important rote in the progress of malignent tumor. The aim of this study is to observe the ability of celecoxib to sensitize five different strains of human malignent cells to radiation in vitro and to prove the possible radiosensitive mechanism. The subtoxic dose of celecoxib was chosen to make the radiosensitive experiments on the human glioma cell lines SHG-44 and 767, the human lung adenocarcinoma cell lines A549, the human cervical carcinoma cell lines HeLa and the human nasopharyngeal carcinoma CNE. The cells were planted into two groups respectively, one was radiation only (R), and the other was radiation combined with 30μmol/L eelecoxib (R+D). Celecoxib's radiosensitization was measured by clongenic assay. Celecoxib displayed radiosensitizing effect in the five strains of malignant cells. The survival curve and the related parameters such as: Do, Dq, SF2 and D0.01in group D+ R decreased significantly from Group R. It is indicated that the subtoxic dose of Celecoxib could enhance radiosensitivity of SHG-44 and 767 cells, A549 cells, HeLa cells and CNE cells in vitro. The radiosensitivity enhencement by the affection of Celecoxib is universal in different malignant tumor cells.
