摘 要:目的:建立HPLC快速测定人血清中普卢利沙星活性代谢物(NM394)浓度。方法:以氟罗沙星为内标,样品经酸化沉淀蛋白后,上清液先后分别以3mI。正己烷和2H止醋酸乙酯进行反洗后进样。分析柱Diamonsil Platisil C18(150mm×4.6mm,5μm),保护柱Dikma EasyGuard C18(8mm×4mm,5μm),柱温35℃;流动相为乙腈-0.02mol·L^-1磷酸二氢钾(pH2.7,含0.5%三乙胺)(10:40.5),流速1mL·min^-2,检测波长275nm。结果:NM394在10~5000μg·L^-1范围内线性关系良好(r^2=0.9997),最低定量限为10μg·L^-1。日内、日间精密度均小于6%,方法学回收率为96.31%~102.49%。结论:本方法快速、准确、灵敏、选择性高,适用于普卢利沙星人体药动学研究。OBJECTIVE To develop an HPLC detection for the quantification of active metabolite of prulifloxacin (NM394) in human serum. METHODS Fleroxacin was used as internal standard (IS), after protein precipitation and a simple two-step back-wash procedure, NM394 and 1S were separated on a C18 column with a mobile phase containing acetonitrile 0. 02 mol· L^-1 KH2 PO4 (pH 2. 7, containing 0. 5 % of triethylamine) (10: 40. 5) at a flow rate of 1 mL·min^-1. The analytes were detected at 275 nm. RESULTS Calibration curves were linear (r^2 = 0. 999 7) from 10-5 000μg·L^-1. Both intra-day and inter-day precisions were less than 6%. Methodology recovery was 96. 31%-102.49%. Limit of quantification for NM394 was 10μg·L^-1. CONCLUSION HPLC described herein had been successfully applied for human pharmacokinetic studies, which offers advantages of specificity,quickness and higher sensitivity over the previously reported methods.
