摘 要:对甘蔗种茎进行检疫时通常需要提取其基因组DNA,而前人有关提取甘蔗DNA的文献报道多是以叶片为材料。为探索从茎汁中提取基因组DNA的有效方法,分别采用CTAB法、SDS-Ⅰ法和SDS-Ⅱ法进行提取,并以叶片为对照。对所提取的DNA质量进行了检测,结果表明:从茎汁中提取DNA时,SDS-Ⅰ法的产率最高,浓度达到235.30ng/μL。整体而言,种茎中提取到的DNA产率不如叶片高,但其浓度和纯度足以满足下游分子实验的要求,且其操作过程较叶片简便。Genomic DNA usually needs to be extracted from sugarcane seed-stem for quarantine.Leaves are mostly used in previous literature reports on this area.Three different methods were taken to extract genomic DNA from sugarcane seed-stem,and the methods were CTAB,SDS-Ⅰand SDS-Ⅱ.Leaves were taken as the control.The quality of the extracted DNA was detected.The results showed that DNA yield of SDS-Ⅰwas the highest.Overall,although the yield of DNA from seed-stem was lower than leaves,but the concentration and purity of DNA from seed-stem were sufficient to meet the requirement of the downstream molecule experiments,and the operation was more convenient than taking leaves as material.
