摘 要:目的:观察附子理中丸对脾阳虚证大鼠适应性产热的增强作用。方法:18窝新生Wistar大鼠,同窝匹配、体重均衡,随机分9组,不切除甲状腺的5组为获得性脾阳虚,切除甲状腺的4组为发育性脾阳虚。d_1切除部分甲状腺,d_(42)切除肩胛骨间棕色脂肪,d_(56)高脂饲养,d_(70)用药防治。每周称量计算体重增长率,d_(98)观察异丙肾上腺素诱导大鼠适应性产热的峰值(℃)和曲线下面积(℃·h),t检验比较组间差异。结果:获得性脾阳虚大鼠,模型组与正常组相比体重增长降低37.1%(P<0.01),适应性产热峰值降低3.2%(P<0.01),曲线下面积减少3.4%(P<0.01)。温治组与高脂组相比,体重增长增加33.6%(P<0.01),适应性产热峰值升高0.5%(P<0.01),曲线下面积增加0.7%(P<0.01)。发育性脾阳虚大鼠,模型组与正常组相比体重增长降低16.6%(P<0.01),适应性产热峰值降低5.5%(P<0.01),曲线下面积减少5.6%(P<0.01)。温治组与高脂组相比,体重增长降低58.8%(P<0.01),适应性产热峰值升高1.1%(P<0.01)、曲线下面积增加0.9%(P<0.01)。结论:附子理中丸增强获得性和发育性脾阳虚模型大鼠的适应性产热。Objective: To observe the effect of Aconitum Lizhong pill on the adaptive thermogenesis in rats with spleen yang defi- cient syndrome. Method: Eighteen litters were used in this experiment. Nine littermates in each litter were bred sequentially and ran- domized into 9 groups. Five groups were not carried out thyroidectomy, the other four groups did. Thyroidectomy in hypothyroidism littermates was carried out with 131iodine to destroy partly thyroidal tissue at d1 after birth, the interscapular brown adipose tissue was removed at d42 ,littermates were fed with more cholesterol ester and triglyceride from d56, then administered intragastrically a decoction of medical herbs to promote or inhibit chronic thermogenesis from d70 to dgs. The body weight were weighed weekly to get the growth ratio, the thermogenesis induced with isoproterenol was evaluated in thermoneutrality (28 ~℃ ) at d98, the peak value ( ~C ) and the area under the curve ( ℃·h) of the core temperature were observed. The potency data were derives from the primary data. A t-test was used to evaluate variation between groups, P 〈 0. 05 as the statistic significant. Result: In the euthyroid rats, compared with the control, the weight growth ratio decreased 37. 1% (P 〈0. 01 ) in model rats,the peak value of the core temperature lowed 3. 2% (P 〈0.01 ) and the area under the curve decreased 3.4% (P 〈 0. 01 ). The weight growth ratio of rats treated with Fuzi-Lizhong Pill improved 33. 6% ( P 〈 0. 01 ) than the fat-diet, and the peak value of the core temperature increased 0. 5 % ( P 〈 0.01 ), the area under the curve ( ℃· h) increased 0.7 % ( P 〈 0.01 ). In hypothyroid rats, compared with the control, the weight growth ratio decreased 16. 6% (P 〈 0. 01 ) in model rats,the peak value lowed 5. 5% (P 〈0. 01 ) and the area under the curve decreased 5.6% (P 〈0.01 ).
