摘　　要：目的:探讨去甲肾上腺素(noradrenalin,NA)对THP-1巨噬细胞ATP结合盒转运体A1(ATP binding cassette transporter A1,ABCA1)、A类清道夫受体(Scavenger receptorAⅠ,SR-AⅠ)和B型清道夫受体CD36(Cluster of Differentiation 36,CD36)表达的影响。方法:不同浓度NA处理THP-1巨噬细胞24小时,运用逆转录多聚酶链反应(reverse transcription polymerase chain reaction,RT-PCR)检测其ABCA1、SR-AⅠ和CD36的mRNA表达,western-blotting检测ABCA1和SR-AⅠ蛋白的表达。结果:100nmol/L,1μmol/L及10μmol/L的NA作用于THP-1巨噬细胞,引起ABCA1 mRNA和蛋白水平下调(P<0.05),1μmol/L及10μmol/L的NA则升高SR-AⅠ mRNA和蛋白的水平(p<0.05),10pmol/L～10μmol/L的NA作用THP-1巨噬细胞24小时,各组CD36 mRNA表达与对照组比较其差异无统计学意义。结论:应激浓度的NA可能通过影响ABCA1和SR-AⅠ的表达而参与和/或促进AS的发生发展。Objective：To investigate the effects of NA on the expression of ABCA1,SR-AⅠ and CD36 in THP-1 macrophages.Methods：THP-1 macrophages were incubated with different concentrations of NA for 24 hours,then the mRNA levels of ABCA1,SR-A Ⅰand CD36 in the THP-1 macrophages were determined by RT-PCR.Western-blotting analysis were used to dectect protein level of ABCA1 and SR-AⅠ.Results：THP-1 macrophages is treated with 100 nmol/L,1 μmol/L and 10 μmol/L of NA,the mRNA and protein levels of ABCA1 decrease（p〈0.05）,then 1 μmol/L and 10 μmol/L noradrenalin cause the mRNA and protein levels of SR-A Ⅰ increase（p〈0.05）.But noradrenalin have no significant effect of CD36 gene expression.Conclusion：Stress concentrations of NA could down-regulate ABCA1 expression and up-regulate SR-AⅠ expression in THP-1 macrophages,which might be related to the genesis of atherosclerosis.
