新疆雪莲愈伤组织诱导及总黄酮的测定

摘　　要：以新疆雪莲植株叶片为外植体,研究了不同植物激素和培养时间对新疆雪莲愈伤组织生长、总黄酮含量和产量的影响。结果表明:新疆雪莲愈伤组织最佳诱导培养基为MS+2.00 mg/L NAA+1.00 mg/L 6-BA,其愈伤组织诱导率最高(97.0%);继代培养基以MS+2.00 mg/L NAA+1.00 mg/L6-BA中愈伤组织生长量和总黄酮产量最高,分别达4.58 g和12.24 mg;以MS+4.00 mg/L NAA+2.00 mg/L 6-BA中的愈伤组织总黄酮含量最高,达2.17 mg/g;愈伤组织的生长量随继代培养时间的延长而增加,而其总黄酮含量不稳定,于继代培养第30天达到最高(2.7 mg/g),此时总黄酮产量最高可达26.66 mg。With the leaves of Saussurea involucrata as explants,the effects of different plant hormone and culture time on the callus growth,total flavonoid content and total flavonoid output of S.involucrata were investigated in this test.The results showed that MS＋2.00 mg/L NAA＋1.00 mg/L 6-BA was the best induction medium,which callus induction rate was the highest（97.0%）.The value of the highest growth and the output of total flavonoids were 4.58 g and 12.24 mg respectively on this medium.The highest content of total flavonoids appeared on the medium with MS＋4.00 mg/L NAA＋2.00 mg/L 6-BA and the value was 2.17 mg/g.Callus growth increased with the culture time prolong,but the content of flavonoids was not stable.The callus total flavonoid content and output reached to the highest after being cultured for 30 days and the values are 2.7 mg/g and 26.66 mg,respectively.
