凡纳滨对虾基因组fosmid文库的构建及钙离子通道基因的筛选

摘　　要：本研究构建了凡纳滨对虾的基因组fosmid文库,该文库包含大约1.1×105个克隆,插入片段平均长度大于35 kb.根据和果蝇钙离子通道基因同源的凡纳滨对虾EST序列设计引物,对文库进行了PCR筛选,得到1个阳性克隆,测序结果显示该克隆包含有钙离子通道基因.研究结果为凡纳滨对虾的基因克隆和分子选育等研究奠定了基础.A fosmid genomic library was constructed for Litopenaeus vannamei.The library consisted of about 1.1×105 clones,in which average length of inserts was greater than 35 kb.The library was screened by PCR analysis using primers designed according to the EST from L.vannamei homology to calcium-release-channel gene of Drosophila melanogaster.A positive clone was selected,subjected to DNA sequencing,and an existence of the calcium-release-channel gene was revealed.These results will improve the study on gene cloning and molecular breeding for L.vannamei.
