摘　　要：目的:观察蜕皮甾酮对实验性大鼠胃溃疡愈合的影响并探讨其作用机制。方法:采用改良Okabe法制备大鼠胃溃疡模型。80只大鼠随机均分为空白对照组(A组)、奥美拉唑钠组(B组)、蜕皮甾酮组(C组)、蜕皮甾酮联合奥美拉唑钠组(D组)。分别于灌胃治疗第7天、第14天后各处死一半大鼠,取组织标本,光镜下观察溃疡形态及愈合情况并测定溃疡指数,免疫组化法检测胃黏膜组织中表皮生长因子(EGF)及表皮生长因子受体(EGFR)的表达情况、酶联免疫吸附试验法检测大鼠胃溃疡周边组织中前列腺素E(2PGE2)的含量。结果:灌胃治疗第7天和第14天,大鼠胃溃疡指数A组>C组>B组>D组(P<0.05)。EGF及EGFR的表达及PGE2的含量均为A组<B组<C组<D组(P<0.05)。结论:蜕皮甾酮可能通过诱导胃组织中EGF、EGFR和PGE2的高表达而促进大鼠胃溃疡的愈合,该作用尤其在与奥美拉唑钠联合应用时更强。OBJECTIVE：To investigate the effect of ecdysterone on the healing of experimental gastric ulcer in rats and its mechanism.METHODS：The ulcer model was prepared by modified Okabe＇s method.80 rats were randomly divided into group A（blank control）,group B（omeprazole sodium）,group C（ecdysterone） and group D（ecdysterone combined with omeprazole sodium）.And half of them were sacrificed to obtain the histological specimen after 7 days and 14 days treatment.The morphology of gastric ulcer and healing condition were observed under light microscope,and ulcer index was determined.The expressions of epidermal growth factor（EGF） and epidermal growth factor receptor（EGFR） were measured by immunohistochemical method.The levels of PGE2 were detected by ELISA method.RESULTS：After 7 days and 14 days of treatment,the descending order of gastric ulcer index was group A,group C,group B,group D（P〈0.05）.The ascending order for the expression of EGF and EGFR and the level of PGE2 was group A,group B,group C,group D（P〈0.05）.CONCLUSION：Ecdysterone can improve the high expression of EGF,EGFR and PGE2 in gastric tissue so as to promote gastric ulcer healing process.Ecdysterone combined with omeprazole sodium shows stronger effect on the healing of gastric ulcer.
