摘  要:将分子印迹技术与表面等离子体共振技术联用,建立了一种新型间接抑制免疫分析法,并用于磺胺甲噁唑的测定。采用管内原位聚合的方法,在长30cm,内径0.25mm的毛细管内制备了磺胺甲噁唑印迹聚合物涂层。涂层厚度由扫描电子显微镜测得为198nm。在BIAcore3000生物传感器上自动进行检测。涂层毛细管用于免疫检测前的在线固相萃取和预富集。磺胺甲噁唑单克隆抗体被富集的磺胺甲噁唑抑制,抑制信号与磺胺甲噁唑的浓度成正比。在优化的实验条件下,方法的线性范围为0.04~10.0μg/L;检出限为0.01μg/L。本方法可直接用于实际样品的检测,回收率好,灵敏度高,操作简便,自动化程度高。A new indirect inhibitive immunoassay using surface plasmon resonance (SPR) coupled with molecularly imprinted polymer (MIP) was developed and applied to the analysis of sulfamethoxazole (SMX).A SMX MIP coating was synthesized in a 30 cm 0.25 mm i.d flexible quartz capillary by in situ polymerization technique.Thickness of the coating was 198 nm illustrated by scanning electron microscope (SEM) image.The detection was conducted on BIAcore 3000 sensor automatically.The MIP coating capillary was used as on-line solid phase extraction (SPE) tube before SPR detection.The anti-SMX mono-antibody was inhibited by SMX that was extracted by the MIP coating in a dose-dependent manner.The calibration curve was obtained by linear fit over the range of 0.04-10.00 μg/L.The detection limit was 0.01 μg/L.It was applied to the analysis of lake water samples.This method has high sensitivity and can be performed automatically.
