摘 要:以无花果曲霉(Aspergillus ficuum 3.4322)基因组DNA为模板,用PCR方法扩增植酸酶基因(phyA)。将PCR产物克隆到pMD 18-T质粒中,用于DNA测序。DNA序列分析结果表明,基因全长1 509 bp,其中包括一个长105 bp的内含子。编码467个氨基酸,第1-19氨基酸为信号肽序列,成熟的植酸酶由448个氨基酸组成。与黑曲霉(Aspergillus niger)植酸酶(GenBank Accession:M94550)的氨基酸同源性为95.7%,与无花果曲霉(Aspergillus ficuum3.324)植酸酶(GenBank Accession:AY013315)的氨基酸同源性为98.9%,显示了植酸酶基因在不同菌株中的差异。A Phytase gene(phyA) from Aspergillus ficuum 3.4322 was cloned by PCR using genomic DNA as template.The PCR products were cloned into pMD18-T vector for sequencing.The sequencing results showed that the full length of phyA is 1509bp with an intron of 105bp.It encodes a protein of 467 amino acids.There is a signal peptide of 19 amino acids at the N-terminus.The mature protein of Phytase from Aspergillus ficuum 3.4322 consists of 448 amino acids.The amino acid homology of Phytase from Aspergillus ficuum 3.4322 is 95.7% and 98.9% compared with Phytase from Aspergillus niger(GenBank Accession: M94550) and Aspergillus ficuum 3.324(GenBank Accession: AY013315),respectively.It shows the diversity of Phytase gene in different strains.
