摘 要:该研究旨在探讨中国重点保护鸭品种催乳素(PRL)基因内含子1和2的单核苷酸多态性(SNP)水平。利用PCR-SSCP和测序的方法检测了8个地方鸭种共476个个体的PRL基因内含子1和内含子2的单核苷酸多态性,分析了不同群体的等位基因频率和基因型频率,并对其遗传特性进行了统计。结果表明:所检测的8个地方鸭种在PRL基因内含子1上均呈现多态,比较测序结果发现位于PRL基因内含子1的383 bp位置有A/C突变;除连城白鸭外,其他各群体中等位基因A均为优势等位基因;除连城白鸭和绍兴鸭外,其他各群体在该位点的基因频率均处于Hardy-Weinberg平衡(P>0.05)。5个鸭种在PRL基因内含子2上存在多态,绍兴鸭、北京鸭和高邮鸭没有发现多态,位于PRL基因内含子2的2 297 bp和2 356 bp位置发生了A/T和G/T的碱基突变;除建昌鸭外,其他鸭群的等位基因D的基因频率较高,而且北京鸭、绍兴鸭和高邮鸭中只发现等位基因D。Hardy-Weinberg平衡适合性检验结果表明,各群体在该位点的基因频率均处于Hardy-Weinberg不平衡状态(P<0.05)。从各个品种的经济类型和群体基因型频率的分布情况来看,PRL基因多态性可能与品种受到的人工选择和自然选择有关。In order to explore the level of single nucleotide polymorphisms(SNP) on intron 1 and intron 2 of prolaction(PRL) gene of ducks in China,the SNP of PRL gene was investigated on eight domestic duck populations of Gaoyou,Jinding,Liancheng White,Putian Black,Beijing,Shaoxing,Youxian Sheldrake and Jianchang,and their gene frequency and genotype frequency were analyzed.The primers for intron 1 and intron 2 of PRL gene were designed according to the database of duck genomic sequence(Genbank no.: AB158611) and the SNPs were detected by PCR-SSCP and sequencing method.The polymorphism(A/C at 383 bp of PRL gene) of intron 1 was found in all the groups,and the frequency of A allele was significantly higher than that of B allele in ducks except for Liancheng White duck.The values of the genotypes of the groups were consistent with the Hardy-Weinberg equilibrium(P〉0.05) except Liancheng White duck and Shaoxing duck.Moreover,the polymorphism(A/T at 2 297 bp and G/T at 2 356 bp of PRL gene) of intron 2 was only found in Jinding duck,Liancheng White duck,Putian Black duck,Youxian Sheldrake duck and Jianchang duck,and the frequency of D allele was higher than that of C allele in ducks except for Jianchan duck and only D allele was detected in Beijing duck,Shaoxin duck and Gaoyou duck.The values of the genotypes of the all groups were not consistent with the Hardy-Weinberg equilibrium(P〈0.05).The gene frequency distribution and economical function of ducks reveal that PRL gene polymorphisms which are significantly associated with duck productivity,are likely related to artificial and natural selection.
