摘 要:目的建立体外培养大鼠脑皮质星形胶质细胞液压冲击损伤模型。方法将30只新生24h内Sprague-Daw-ley(SD)大鼠脑皮质制成单细胞悬液后原代培养于8个玻璃培养瓶,经纯化后传代于80个培养皿中,随机分为5组,各16个;以不同的液压损伤细胞0.5h后,光镜下观察各组细胞形态学变化,Hoechst33342/PI荧光染色和乳酸脱氢酶(lactate dehydro-genase,LDH)检测细胞的损伤程度。结果体外培养星形胶质细胞生长状态良好,纯度达(94.9%±1.2%);不同大小的冲击压力可导致细胞不同程度的损伤,形态学变化明显;随冲击压力增大,红色荧光细胞数量(P<0.01)和LDH释放量逐渐增高(P<0.01)。结论经改良的细胞液压冲击损伤模型是体外研究创伤性损伤的有效模型。Objective To develop a model of hydraulic impact injury in primary cultured rat astrocytes in vitro. Methods Cell suspension of rat cerebral cortex from 30 Sprague - Dawley rats were cultured primarily in eight glass culture bottles, and then were cultured secondarily in 80 petri dishes. The petri dishes were randomly divided into five groups. After half an hour of different fluid impact pressures, the morphological changes of astrocytes were observed under light microscope, the extent of cell injury was qualitatively assessed by Hoechst33342/PI fluorescence staining and lactate dehydrogenase (LDH) enzyme release. Results The cultured astro- cytes grew well, and their purity reached 94.9% ±1.2%. The different degrees of cell injury were caused by different fluid impact pressures. With the increase of the pressure, the quantity of red fluorescent cells (P 〈 0.01 ) and the value of LDH also increased (P 〈0.01 ). Conclusions It is a useful model to study traumatic injury in cultured cells.
