摘  要:为探讨细胞间粘附分子-1(ICAM-1)在着床中的作用和调节,本研究采用昆明种小鼠,获取孕第一天的单细胞胚胎,分别在M16培养液和含0.1ng/ml浓度白血病抑制因子(LIF)的M16培养液中培养120小时,每24小时更换培养液,然后,用免疫印迹法测定胚胎培养液中的ICAM-1在蛋白水平的表达。结果:(1)5天的培养液中均有ICAM-1的表达。第一天培养液中的ICAM-1水平稍高,第二天最低,以后逐渐升高,到第五天达最高峰。(2)LIF可显著提高胚胎ICAM-1的表达水平。结果表明:小鼠孕早期胚胎可以表达ICAM-1,其表达水平在胚泡期最高。ICAM-1的表达可能与着床过程胚泡的粘附有关。细胞因子LIF可促进ICAM-1的表达。Objective:To study the expression of intercellullar adhesion molecule 1(ICAM 1) and the effect of leukaemia inhibitory factor (LIF) on the expression of ICAM 1 in the early embryo. Design: An experiment study with mouse embryos in vitro. Setting:Institute of Obstetrics and Gynecology Hospital,Institute of Liver Cancer of Zhongshan Hospital,Shanghai Medical University. Methods: Single cell embryos were incubated in M16 medium or M16 containing,0.1 ng/ml LIF for 120 hours.The culture medium was changed each day and the protein level of ICAM 1 in the medium was measured with immunoblotting method. Results:(1)The level of ICAM 1 was higher on the first day than on the second.The level started to increase from the second day,reaching the highest level at the fifth day.(2)LIF improved the expression of ICAM 1 in the embryos. Conclusion:ICAM 1 was expressed in the early embryos of mouse.The level of ICAM 1 is the highest in the stage of blastocyst than in other stage.The expression of ICAM 1 may be related to the attachment of blastocyst during implantation.The expression of ICAM 1 in the embryos can be improved by LIF.
