豚鼠耳蜗一氧化氮合酶组织化学反应的定量观察

摘  要:应用NADPH—d酶组化方法结合显微图像分析技术,定量观察了正常豚鼠耳蜗NOS的分布及活性。结果表明:NOS存在于豚鼠外周听觉系统,分布部位依其活性强弱依次为血管纹、内毛细胞、螺旋神经节、外毛细胞、听神经纤维,其相应部位的NOS阳性反应物的平均密度值为93.92%、91.17%、84.25%、78.75%、32.92%,平均灰度值为66.18、73.42、76.67、85.17、111.33。The distribution of NO synthase (NOS) was quantitatively studied by means of NADPH-diaphorase histochemistry staining combined with image quantitative analysis technique in normal guinea pig cochlea. The results showed that the NO synthase was present in the peripheral auditory system. Based on extent of NO synthase activity, the region of distribution in turn were stria vascularis, inner hair cells, outer hair cells , spiral ganglion cells and auditory nerve fibers. The average densities of above regions were 93.92%, 91.71%, 84.25%, 78.75% and 32.92% respectively, while the average grey level of above regions were 66.18, 73.42, 76.67, 85.17 and 111.33 respectively. The results suggest that NO in the peripheral auditory system is related to the auditory modulation and might play an improtant role in the physiology and pathophysiology of the mammalian cochlea. The characteristics and the physiological significance of NO synthase distribution in the cochlea were discaussed.
