摘  要:应用电离辐射诱导人类T淋巴细胞白血病Jurkat细胞株,再经二维凝胶电泳分离不同照射剂量的细胞总蛋白,研究电离辐射诱导的人类T淋巴细胞白血病细胞蛋白的差异表达.应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)进行处理,得到肽质量指纹(PMF)图谱,确认6个差异表达蛋白点.Two-dimensional electrophoresis and matrix-assisted laser desorption/ionization time-of-flight mass spectrorometry(MALDI-TOF-MS) have been acknowledged as the efficient methods in proteomics studies.In this report,the total proteins of human T lymphocytic leukemia(Jurkat cells) treated by ionizing radiation were separated with two-dimensional electrophoresis.The differential expression of cellular proteins induced by ionizing radiation was analyzed by PDQUEST software.Six differential protein spots were identified by MALDI-TOF-MS,including Lamin B1,heterogeneous nuclear ribonucleoprotein,p47 protein isoform a,alternative splicing factor 1,prohibitin,and 6-phosphogluconolactonase.The results may help researchers better understand the underlying mechanism involved in cellular protein alteration induced by ionizing radiation.
