摘 要:目的:建立梯度洗脱测定肾康灵颗粒中丹参素、原儿茶醛含量的HPLC法。方法:Hypersil ODSCl8柱,流动相甲醇-1%冰醋酸溶液,线性梯度洗脱:0~8 min(5∶95),8~11 min(6∶94~5∶95),11~20 min(5∶95);流速1 mL.min-1;检测波长280 nm。结果:在此色谱条件下2种成分可完全分离。丹参素、原儿茶醛的线性范围分别为0.050 4~0.504μg(r=0.999 8),0.109 0~1.090μg(r=0.999 9)。丹参素、原儿茶醛在肾康灵颗粒中的平均回收率分别为98.40%,98.85%,RSD 1.2%,0.87%。结论:该方法简便、准确、快速,可同时测定肾康灵颗粒中丹参素、原儿茶醛的含量。Objective: To develop an HPLC method for gradient elution determination of danshensu and protocatechuic aldehyde in Shengkangling granula. Method: The analysis was carried on a column of Hypersil ODS2-Cls with a mobile phase consisted of methanol-1% glacial acetic acid 0-8 min(5: 95), 8-11 min(6: 9±5:95), 11-20 min(5: 95) gradient elution at the flow rate of 1 mL·min^-1. The detection wavelength was 280 nm. Result: The linearity was obtained in the range of 0. 050±0. 504μg( r = 0. 999 8 ) for tanshinol and 0. 109 0-1. 090 μg( r = 0. 999 9)for protocatechuic aldehyde, respectively. The average recoveries of tanshinol and protocatechuic aldehyde in Shenkangling granula were 98.40% and 98.85% , and the RSDs were all less than 2.0%. Conclusion: The method is simple, accurate and rapid. It may be suitable for the usual quality control of tanshinol and protocatechuic aldehyde in Shengkangling granula.
