老年性耳聋患者内皮祖细胞增殖和衰老变化及其意义

摘　　要：目的探讨体外培养的老年性耳聋患者外周血来源的内皮祖细胞的增殖和衰老变化情况及其临床意义。方法测定培养7d的老年性耳聋患者和对照组健康老年人内皮祖细胞增殖能力、细胞集落数,比对两组培养14d时内皮祖细胞的衰老程度。结果老年性耳聋组内皮祖细胞增殖能力为(0.154±0.042),对照组为(0.226±0.574),两者比较差异具有统计学意义(t=3.209,P=0.005<0.01);内皮祖细胞计数:老年性耳聋组为(54.32±9.24)个,对照组为(67.64±8.39)个,两者比较差异具有统计学意义(t=3.376,P=0.003<0.01);内皮祖细胞集落计数:老年性耳聋组为(1.40±0.37)个,对照组为(2.88±0.48)个,两组相比差异具有统计学意义(t=7.734,P<0.01);衰老细胞计数为:老年性耳聋组为(43.90±4.11)个,对照组为(39.76±4.01)个,两组相比差异具有统计学意义(t=2.280,P=0.035<0.05)。结论老年性耳聋患者内皮祖细胞增殖能力下降,易于衰老,可能是老年性耳聋血管退化的机理之一。【Objectives】To investigate the proliferative function and senescence of endothelia progenitor cells（EPC） from peripheral blood in patients with presbycusis,and explore the implication to presbycusis.【Methods】 At 7 day,the proliferation of EPC Numbers of EPC colonies and EPC were investigated and comparison was made between presbycusis group and the healthy aged group.At 14 day,the senescence of EPC were investigated and compared between two groups.【Results】The proliferative faculty of EPC decreased significantly in presbycusis group compared to contrast group（0.154 ± 0.042 versus 0.226 ± 0.574,P 0.01）,the number of EPC（54.32 ± 9.24 versus 67.64±8.39,P 0.01）,EPC colonies（1.40±0.37 versus 2.88 ± 0.48,P 0.01） also decreased significantly respectively.Compared to contrast group,the senescence in presbycusis group was increased significantly（43.90 ± 4.11 versus 39.76 ± 4.01,P 0.05）.【Conclusion】The proliferative faculty of EPC significantly decreased,senescenced of EPC increase significantly in patients with presbycusis.This may be one of the mechanisms on blood capillary degeneration of inner ear in presbycusis.
