摘 要:目的:通过调查大鼠脑干损伤组织的S100B-mRNA与S100B的表达特点,系列了解损伤后不同时间点脑干组织S100B表达的变化规律并分析其与损伤时间的关系。方法:采用撞击法制作实验大鼠脑干损伤动物模型,采集损伤后2~48h内4个不同时间点的脑干组织,实时荧光定量PCR方法检测大鼠脑干损伤后S100B-mRNA的表达变化,免疫组化(SP)法观察S100B不同时间点的表达变化。结果:(1)伤后2h组S100B-mRNA表达下降,之后随时间的延长不断升高,至24h达到高峰,随后下降,至48h时基本降至正常水平。(2)伤后2hS100B表达增多,并逐渐升高,至24h达到高峰,随后下降,但至48h仍高于正常水平。结论:原发性脑干损伤后S100B-mRNA及S100B表达变化有一定的规律,对脑干损伤的致伤时间推断具有参考价值。Objective To investigate S100B expression in rat brainstem and the relationship between S100B expression and the injury period after primary injury. Methods The brainstem injury animal model was established using the impaction method, the injured brainstem tissues were collected at various time points within 2 to 48 hours. Real-time PCR was performed to detect S100B mRNA expression, and immunohistochemistry (SP) was used to detect S100B protein expression at different time points after injury. Results S100B mRNA expression was decreased in the 2 h injury group and was elevated in time-dependent manner until 24 h after injury, and was decreased to normal level at 48 h after injury. However, S100B protein was increased during the period from 2 h to 24 h and then decreased at 48 h after injury, but, S100B expression was still higher than that of the normal level at 48 h after injury. Conclusion S100B mRNA and protein expression in rat brainstem was regulated after primary injury, which can indicate the real time points for the brainstem injury.
