摘  要:目的:探讨低氧预适应(HPC)对肢体缺血-再灌注(I/R)大鼠肝脏的保护作用。方法:重复性低氧5次建立大鼠HPC模型,以橡皮带环绕结扎大鼠双后肢跟部,阻断血流4h后再灌注4h复制肢体I/R模型,设立对照组、I/R组和HPC+I/R组。检测各组血中天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、乳酸脱氢酶(LDH),血中和肝组织中超氧化物歧化酶(SOD)、黄嘌呤氧化酶(XOD)、丙二醛(MDA)的含量变化及肝组织的湿/干重比值(W/D)。结果:I/R组大鼠血中ALT、AST、LDH含量升高;血中和肝组织中XOD、MDA含量升高;血中和肝组织中SOD活性降低;肝组织的W/D增高。HPC+I/R组大鼠上述指标较I/R组均明显改善,差异有显著性。结论:低氧预适应对肢体缺血/再灌注后肝脏的损伤有明显的保护作用。Objective:To investigate the effects of hypoxic preconditioning(HPC) on liver function in limb ischemia/reperfusion(I/R) injure rats.Methods:HPC model was established for 5 times of repetitive hypoxia.Rats were randomly divided into three groups:control group,I/R group and HPC+I/R group.I/R injury model rats underwent 4h of limb ligation followed by 4h reperfusion.The values of alaninea minotransferase (ALT),aspartate aminotransferase (AST) and lactate dehydrogenase(LDH) in the plasma were observed,the contents of superoxide dismutase(SOD),xanthine oxidase(XOD),malondialdehyde(MDA) in the plasma and the liver tissue were measured,the ratio of wet/dry(W/D) weight of liver tissue was measured.Results:The values of ALT,AST and LDH were significantly higher than those of control group in the plasma;the contents of XOD and MDA were higher in the plasma and liver tissue;the activity of SOD was significantly lower in the plasma and liver tissue;the ratio of wet/dry(W/D) weight of liver tissue was higher.In HPC+I/R group,that rats subjected to HPC,the liver function were significantly improved when rats exposed to limb I/R injury.Conclusion:Hypoxic preconditioning in rats can protect liver ofter limb I/R injury.
