抗IL-14R单抗在SLE患者中对IL-14R的表达及其Ig分泌的影响

摘　　要：目的探索ＩＬ１４Ｒ与ＳＬＥ病程的相关性。方法应用抗ＩＬ１４Ｒ单抗（ＢＡ５）分析了３０例正常人和４３例ＳＬＥ患者外周血淋巴细胞表面ＩＬ１４Ｒ的分布与表达。结果发现ＳＬＥ患者外周血ＩＬ１４Ｒ阳性细胞百分比（２２５６％）明显高于正常人（７８４％）。在活动期和缓解期ＳＬＥ患者中，ＩＬ１４Ｒ表达百分比也具有显著的差异（３２０２％和１３０９％）。结论提示ＢＡ５单抗能拮抗Ｂ细胞激活后ＩＬ１４Ｒ的高表达和ＩｇＧ的分泌。并可能以ＢＡ５单抗阻断和调节ＩＬ１４Ｒ的表达等方面调节Ｂ细胞功能的异常，从而为临床提供新的免疫治疗和检测手段。Objective To explore the IL 14R expression in peripheral blood of SLE patients and to analyse the relationship between IL 14R and course of disease.Methods IL 14R positive subset in PBMCs of 30 healthy subjects and 43 SLE patients was determined by flow cytometry with BA5,an mAb anti IL 14.Results Significant difference of the percentage of IL 14R positive cells was detected not only between SLE patients (22 56%) and healthy subjects (7 84%),but also between the patients in active (32 02%) and in remission periods (13 09%). Moreover,an IL 14 containing supernant of mixed lymphocyte culture (MLRS) in the experiment to induce a more intensive proliteration and a higher expression of IL 14R in the SAC stimulated B cells and PBMCs for both patients and healthy subjects.The mAb BA5,however,was capable of blocking the MLRS' effect,resulting in a derease of B cells′ relative response from 100% to 40 18% and 44 70% for healthy subjects and patients,respectively.Conclusion These results show a promise for immunoprevention with mAb BA5 for control of the autoimmune diseases.
