摘  要:目的建立白斑颗粒中川芎嗪的提取和含量测定方法。方法先用盐酸-乙醇溶液提取样品,D-101大孔吸附树脂富集纯化盐酸川芎嗪,再用反相高效液相色谱法测定含量。色谱柱为Inertsil ODS-3柱(150mm×4.6mm,5μm),以甲醇-0.05mol/L柠檬酸溶液(30:70,V/V)为流动相,检测波长300nm,流速1.0mL/min,柱温30℃,进样量10μL。结果盐酸川芎嗪质量浓度在5.76~13.44μg/mL范围内与峰面积线性关系良好(r=0.9999),样品的平均回收率为96.79%,RSD=1.78%。结论所建立的提取分离方法能较好地提取分离盐酸川芎嗪;反相高效液相色谱法专属性较强,能准确测定白斑颗粒中盐酸川芎嗪含量。Objective To establish the method of extracting, separating and determining the content of ligustrazine hydrochlofide in Baiban Granules. Methods Under the use of hydrochloric acid -ethynol solution for extracting and D- 101 macroporous adsorbed resin for saparating, ligustrazine hydrochloride was determined by RP- HPLC. HPLC was performed on the Inertsil ODS- 3 column(150 mm× 4. 6 ram, 5 μm). The mobile phase consisted of methanol -0.05 mol/L citric acid (30 : 70, V/ V). The flow rate was 1.0 mL/min. The detection wavelength was 300 nm. The column temperature was 30℃. The injection volume was 10μL. Results The concentration linear range of ligustrazine hydrochloride was 5.76- 13. 44μg/mL( r=0. 999 9) with the recovery rate of 96. 79%. Conclusion The established method can conduct the better extraction and separation of ligustrazine hydrochloride, which can accurately measure ligustrazine hydrochloride in Baiban Granules with strong specificity.
