摘 要:比较人类正常体细胞染色体核型和食管癌细胞系EC-1和EC-109染色体核型,了解食管癌细胞系EC-1和EC-109的细胞遗传特征,为研究食管癌的癌变机理提供了方向.利用秋水仙素、KCl低渗液处理细胞,经卡诺氏固定液固定制备染色体,滴片,进行Giemsa染色、镜检、选取染色体长度适中且分散程度好的染色体拍照,采用Adobe Photoshop软件进行染色体核型分析.结果表明:与正常人类细胞比较,食管癌鳞状上皮细胞系EC-1和EC-109的染色体核型出现明显异常,染色体核型分析为亚三倍体,众数为46~68条.食管癌鳞状上皮细胞系EC-1和EC-109的细胞遗传特征有显著的异常改变,细胞恶性转化特质明显.它们是研究癌变机理的良好细胞模型,可用于研究CNV与癌变的关系等.Understanding the cytogenetie characterization of EC-1 and EC-109 may provide a new direction for the study of carcinogenesis mechanism of esophageal cancer by comparing the karyotypes of esophageal carcinoma celllines of EC-1, EC-109 with the normal somatic cells. EC-1 and EC-109 cells in logarithmic growth phase were treated with eolchicines, hypotonie solution, Carnoyrs Fluid and Giemsa staining in turns. By observing the chromosomes under the oil lens of microscope,photographs of the chromosomes with moderate length and good dispersion were taken , and analyzed with Adobe photoshop. There were obvious differences on chromosome karyotypes among the esophageal carcinoma cell-line EC-1, EC-109 and the normal somatic cells. Karyotype analysis showed that chromosomes in EC-1 and EC-109 cells were hypotriploidy of modal number 46-68. The molecular and cytogenetic characterization of EC-1 and EC-109 were obviously changed, and the cells malignant conversion appeared. The large fragment aberrations of chromosomes provide a new way to study carcinogenesis mechanism of esophageal cancer, i. e. the relationships between genomic CNV (copy number variant) and carcinogenesis mechanism.
