摘　　要：目的:研究GnRHⅡ对子宫内膜异位症(内异症)患者离体培养的异位及在位子宫内膜间质细胞凋亡的影响。方法:体外原代培养人内异症异位及在位子宫内膜间质细胞,用不同浓度的GnRHⅡ(0、10-10、10-8、10-6mol/L)干预,采用Hoechst染色和流式细胞术检测离体培养子宫内膜间质细胞的凋亡发生情况。结果:GnRHⅡ可诱导离体培养的在位或异位子宫内膜间质细胞的凋亡,随着浓度增加,差异有统计学意义(P<0.05),呈剂量依赖性;且对异位内膜间质细胞的促凋亡作用明显强于在位者(P<0.05)。结论:外源性GnRHⅡ可明显促进人内异症异位及在位子宫内膜间质细胞的凋亡,为开发内异症促凋亡治疗方面的新药提供了实验依据。Objective：To investigate the effect of GnRH Ⅱ on the cell apoptosis of ectopic and eutopic endometrial stromal cells from women with endometriosis in vitro. Methods： The ectopic and eutopic endometrial stromal cells were isolated and cultured in vitro, then exposed to different concentration of GnRH Ⅱ（0,10^-10, 10^-8 , 10^-6mol/L）. The cell apoptosis of ectopic and eutopic endometrial stromal cells were measured by Hoechst staining and flow cy- tometry test. Results：GnRH Ⅱ can increase the apoptotic rate of both ectopic and eutopic cells in a dose-dependent manner（P〈0.05 ） , and the apoptotic rate of ectopic stromal cells was obviously higher than that of the eutopic cells （P〈0.05）. Conclusion： GnRH Ⅱ can significantly enhance the apoptosis of ectopic and eutopic endometrial stromal cells from women with cndometriosis,which will provide the evidence for researching endometriosis drug treatment.
