摘 要:目的研究神经干细胞(neural stem cells,NSCs)在颅脑创伤(traumatic brain injury,TBI)大鼠脑皮层中的转归,及NSCs移植对TBI引起的免疫反应的作用。方法将NSCs移植入TBI大鼠脑皮层,2周后评价NSCs的存活、迁移及分化,并检测脑皮层促炎症因子水平及脑组织炎症反应。结果移植后2周,NSCs在TBI脑皮层中存活,并向病变迁移,TBI组中的迁移细胞是脑组织(1246±92)个/mm2细胞,多数迁移细胞(51.3%±7.5%)具有神经元表型,NSCs移植明显提高TBI大鼠神经运动功能评分,降低急性期促炎症因子水平,并且降低CD3+T细胞及Mac3+细胞数目,从而减轻炎症反应。结论 TBI大鼠脑组织影响NSCs的存活、迁移及分化,NSCs移植的神经保护作用与减轻脑组织炎症反应有一定关系。Objective To investigate the consequence of human neural stem cells(NSCs) after transplantation into the traumatic brain injury(TBI) cortex of rats and examine the changes of TBI induced inflammatory process in transplanted animals.Methods NSCs were transplanted into the injured cortex of TBI rats.We studied the NSCs survival,migration and differentiation 2 weeks after transplantation.We also checked the brain inflammatory process and examined the proinflammatory cytokine levels.Results Transplanted neurospheres survived in TBI brains 2 weeks posttransplantion.Targeted migration of the human cells was seen in TBI rats and many human cells migrated toward the lesion.The migration NSCs of NSCs group were 1246±92 cells/mm2.The majority of migrating cells(51.3%±7.5%) exhibited a neuronal phenotype.NSCs transplantation significantly increased the neurological motor score of TBI rats and down-regulated the inflammatory brain process at the acute phase of disease,as indicated by attenuation of trauma-induced increase in proinflammatory cytokine levels,and reduction in the number of CD3+ T cells and Mac3+ cells.Conclusions Transplanted human NSCs can survive in TBI brains and the microenvironment influences their migration and differentiation.The beneficial effect of transplanted NSCs in TBI brain is mediated,in part,by decreasing brain inflammation and reducing tissue injury.
