摘 要:为了进一步从分子生物学角度探讨首乌延缓衰老的机理,用Norther杂交和Westem印迹法观察了首乌对老年小鼠脾组织中P_(53)基因转录产物mRNA相对含量及翻译产物P_(53)蛋白相对含量的变化的影响。结果表明小鼠脾组织中P_(53)基因的mRNA相对含量及蛋白质相对含量随增龄而增多,用药组mRNA相对合量较老年组下降4.9倍,接近青年组水平,P_(53)蛋白表达电泳显示较老年组明显变浅。为首乌补肾益精、延缓衰老作用提示了分子生物学依据。In order to approach to mechanism of the antisenil function Radix polygorU mndtiflori from molecular biology, applying Norther cross and westere blotting methods to observe the influence of Radix polygoni multiqlori on antiocogent P53 expression in spleen of senile rat. The results showed that mRNA relative content of P53 gene transcriptional product and translational product P53 protein increased as the age of rat increased, the mRNA relative content of P53 gene transcriptional product decreased significantly after taking radix polygon, multiflori, and approached on the level of youth, expression of P53 protein was obviously lighter which showed by electrophoresis than that of senile rat. All these provided with molecular biologic basis for the antisenil function of radix polygon, multiflor.
