淋巴管和瓣膜的动态观察及其与一氧化氮合酶(NOS)关系的实验研究  被引量:1

摘  要:目的研究大鼠肠系膜淋巴管运动及其瓣膜的动态特点;探讨NO对淋巴管的作用及NO在淋巴管及其支配神经的分布。方法用倒置显微镜和图像处理系统对肠系膜及其瓣膜进行动态观察和检测,并取肠系膜淋巴管、胸中段脊髓、延髓、脊神经节和腹腔神经节行NADPS-d组化染色。NADPH-d染色的肠乐膜淋巴管作电镜观察。结果淋巴管收缩频率5~12次,平均为8.7±0.3次。淋巴管运动时瓣膜下游的淋巴管收缩幅度较瓣膜上游的淋巴管收缩幅度大 27.6%。施加 L-Arg后淋巴管口径由2.49±1. 29象素增加到 3. 22± 1.18象素,淋巴管舒张 36.1%。淋巴管管径与两瓣膜间距离的相关关系r=0.297,呈低度相关。瓣膜的关闭和开放与淋巴管的收缩和舒张无明显规律,淋巴液的流动不因淋巴管的收缩而单方向流动。肠系膜淋巴管和瓣膜的内皮细胞以及其周围和附壁的神经纤维皆呈NOS阳性反应。迷走神经背核、胸中部脊髓侧角、腹腔神经节和脊神经节神经元的胞浆及突起皆为NOS阳性反应。电镜下淋巴管内皮细胞胞浆内NOS反应产物,为致密颗粒构成的大小和形状不一的块状结构。结论实验结果提示瓣膜的开放和关闭除了机械因素外可能有其他调节因素。NO可舒张淋巴管,NO既是?Purpose To study the motions of the lymph vessels and their valves in rat mesentery and No effect on the lymph vessels;histochemical localization of nitric oxide synthase in lymph vesseles and their nerve regulation. Method Using inverted microscopy and digital image processing to observe and measured the diameters of the rat mesenteric lymph vessels and the opening and closing of the valves. Using histochemical method and TEM to investigate the localization of NADPH-dependent diaphorase activity within lymph vessels and nerve which innervate the vessels. Results The frequency of automatic motion of the lymph vessels is 8. 7±0. 3 time/min. The correlation between the diameter of the lymph vessels and the gap of the two valves is low. The opening or closing of the valves is not definitely related to the contraction and dilation of the lymph vessels and the direction of the lymph flow is not only one way.After L-Arg was given the lymph vessels dilated about 36. 1 % (from 2. 44±1.29 pixel incresed to 3. 22±1. 88 pixel).Specific marker for nitric oxide synthase was found in endothelia of the lymph vessels and the nerve fibers surrounding the lymph vessels of the rat mesenteries,the neurons of the dorsal nucleus of the vagus nerve and the lateral horns,posterior horn of the spinal cord,the cells of the spinal ganglion and celiac ganglion. Conclusion The results suggested NO dialated the lymph vessels. The opening and closing of the valves may be affected by other factors apart from the mechanic factors. The histochemical localization suggested that nitric oxide may play a relaxation role as neurotransmitters in lymphatic system and also a endogenic relaxation factor.
