肾络康煎剂对实验性IgA肾病模型大鼠TNF-α蛋白表达的影响

摘  要:目的:探讨肾络康煎剂治疗IgA肾病(IgAN)的作用机理。方法:采用脂多糖(LPS)+牛血清白蛋白(BSA)+四氯化碳(CCl4)作为造模方法制作IgA肾病大鼠模型,并分别给予肾络康煎剂和贝纳普利治疗,最后用免疫组化的方法观察各组大鼠肾组织TNF-α蛋白表达。结果:模型组大鼠肾小球IgA荧光较正常,显著组增强(u=3.464 1,P<0.01),而肾络康煎剂组较模型组明显减弱(u=2.327 9,P<0.05);简络康组肾小球系膜细胞数43.75±9.68与模型且62.53±15.33比较(t=-2.929 8,P<0.05),差异显著;肾络康组TNF-α阳性面积(%)为31.25±11.67,较模型组60.67±13.27显著降低(t=-4.708 8,P<0.01)。结论:肾络康煎剂抑制IgA肾病模型肾组织TNF-α蛋白表达可能是其治疗IgA肾病的作用机制之一。Objective: To investigate the mechanism of action of Decoction of Shen - luo - kang on IgA nephropathy. Method: The models of IgA nephropathy were made with lipopolysaccharide, bovine serum albumin and carbon tetrachloride ( CCl4). Respectively Decoction of Shen - luo - kang and benazepril were given to the models. At last TNF - α albumen expression in nephridial tissue in each group was observed with immunohistochemical method. Resuh:IgA fluorescence in rat's glomcrulus in model group was strengthened than that in normal group(u =3.464 1 ,P 〈 0. 01 ), but was weakened obviously in treatment group than that in model group(u = 2. 327 9, P 〈 0. 05 ). Mesangial cell number in treatment group was (43.75 ± 9.68 ), and (62. 53 ± 15.33 )in model group. The difference was significant( t = -2. 929 8, P 〈 0. 05 ). TNF-α positive area in nephridial tissue inhibited by Decoction of shen - luo - kang was ( 31.25 ± 11.67 ) , lower than that in control group which was ( 60. 67 ± 13.2) , (t = - 4. 708 8,P 〈 0. 01 ). Conclusion : Decoction of Shen - luo - kang can inhibit TNF - α albumen expression in nephridial tissue in the models of IgA nephropathy thus to decrease urine protein. It is one of effective treatment methods.
