不同时辰电针足三里对急性胃黏膜损伤大鼠PGE_2、EGF、TGF-α的影响

摘　　要：目的研究前列腺素E2(PGE2)、表皮生长因子(EGF)和转化生长因子α(TGF-α)在大鼠急性胃黏膜损伤中的作用,并探讨不同时辰与胃黏膜保护因子之间的相关性。方法 40只SD大鼠随机分为4组,即空白对照组、模型对照组、辰时电针组、戌时电针组,每组10只。采用无水乙醇灌胃法制成急性胃黏膜损伤模型,用ELISA法检测大鼠血清与胃黏膜中PGE2的含量,并用免疫组化法检测大鼠胃黏膜上EGF与TGF-α表达变化。结果大鼠血浆PGE2与胃黏膜PGE2、EGF、TGF-α水平下降;辰时及戌时电针组电针4d后血浆PGE2与胃黏膜PGE2、EGF、TGF-α均水平升高,戌时电针组血浆中PGE2、EGF、TGF-α表达增高(P<0.01)。结论急性胃黏膜损伤病变与内源性PGE2、EGF和TGF-α的协同保护作用有关,选择合适时辰治疗可提高临床治疗效果和指导评价预后。Objective To study the action of prostaglandin E2（PGE2）,epidermal growth factor（EGF）and transforming growth factor α（TGF-α）in the rats with acute gastric mucosal injury and explore the relativity between time acupuncture with the protective factors of gastric mucosa.Methods 40 SD rats were randomized into 4 groups,named blank control group,model control group,electroacupuncture（7：00am-9：00am）group and electroacupuncture（7：00pm-9：00pm）group,10 rats in each group.Absolute ethyl alcohol was used for gastric perfusion to prepare the model of acute gastric mucosal injury.ELISA assay was adopted to determine the contents of PGE2 in serum and gastric mucosa.IHC method was used to determine the changes in EFG and TGF-α expression in gastric mucosa.Results The levels of plasma PGE2 and gastric mucosa PGE2,EGF and TGF-α all decreased in rats.4 days after electric acupuncture,the levels of plasma PGE2 and gastric mucosa PGE2,EGF and TGF-α all increased in electroacupuncture（7：00am-9：00am）and electroacupuncture（7：00pm-9：00pm）groups.The expressions of gastric mucosa PGE2,EGF and TGF-α in electroacupuncture（7：00pm-9：00pm）group turned higher（P0.01）.Conclusion The acute gastric mucosal injury is relevant with the coordinative protection of endogenous PGE2,EGF and TGF-α.The treatment at an appropriate time division can improve clinical therapeutic effect and guide the evaluation of prognosis.
