摘 要:试验分析比较了3种破壁法和4种提取液法对小麦秆锈菌夏孢子DNA的提取效果。结果表明,玻璃珠液氮振荡法获得的DNA纯度及浓度最高,RAPD扩增条带清晰,为提取DNA的最佳破壁法;CTAB法获得的DNA纯度及浓度最高,满足RAPD扩增条件,条带清晰,为最佳提取液法。适合于小麦秆锈菌RAPD分子标记研究的DNA提取方法的最佳体系为玻璃珠液氮振荡法+CTAB提取液法。In this paper, the different methods including 3 breaking cell wall methods and 4 extraction buffer methods for extracting DNA from wheat stem rust urediospores were compared. The results revealed that the best way for cell wall de- composition was glass bead oscillation method and considerable pure and concentrated DNA and related RAPD bands were obtained. The relatively best quality DNA which met the RAPD analysis demard was obtained by using CTAB method. In a conclusion, the optimal system for DNA extraction was the combination of glass beads oscillation and CTAB extraction.
