抵当汤对去卵巢大鼠血脂异常及ET-1 mRNA、VCAM-1 mRNA表达的影响

摘　　要：目的观察抵当汤对去卵巢大鼠血脂及内皮素1(ET-1)mRNA、血管内皮细胞黏附分子1(VCAM-1)mRNA表达的影响。方法将40只雌性SD大鼠随机分为4组,即假手术组、模型组、抵当汤组和乙烯雌酚组,每组10只。采用手术切除双侧卵巢法复制围绝经期血脂异常的模型,造模2周后假手术组和模型组均给予蒸馏水灌胃,抵当汤组给予含2.5g生药的抵当汤混悬液灌胃,乙烯雌酚组给予乙烯雌酚悬浊液灌胃,均每日1次。连续灌服12周后测定血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)含量及主动脉组织中ET-1mRNA、VCMA-1mRNA表达水平。结果治疗后抵当汤组血清TC、TG及LDL-C水平较模型组降低(P<0.05),HDL-C较模型组升高(P<0.05),与乙烯雌酚组比较差异均无统计学意义(P>0.05);治疗后抵当汤组主动脉组织ET-1mRNA、VCMA-1mRNA表达较模型组降低(P<0.01),与乙烯雌酚组比较差异均无统计学意义(P>0.05)。结论抵当汤可防治去卵巢大鼠血脂异常,具有调节血脂、保护内皮功能的作用。Objective To observe the effect of different dose Didang decoction on blood lipid, the expression of ET-1 mRNA and VCAM-1 mRNA in ovariectomized rats. Methods 40 male rats were randomly divided into four groups, Sham group, model group, Didang decoction and Diethylstilbestrol group. Rat model with climacteric lipid abnormality were duplicated by ovariectomy. Distilled water, Dedang decoction, Diethylstilbestrol suspension were given rats in corresponding groups. After 12 weeks, TC, TG, LDL-C, HDL-C and expression of ET-1 mRNA, VCMA-1 mRNA in aorta tissue were measured. Results The contents of TC, TG and LDL-C in Didang decoction were decreased compared with those in model group （P〈0.05） after treatment. HDL-C was increased （P〈0.05）. There was no significant difference between Didang decoction and Diethylstilbestrol group on the contents of TC, TG and LDL-C （P〉0.05）. The expression of ET-1 mRNA, VCMA-1 mRNA in aorta tissue in Didang decoction were decreased compared with those in model group （P〈0.05） after treatment. Conclusion Didang decoction can regulate blood lipid and protect endothelial function.
