摘 要:目的通过对脑内类氨基酸含量的及相应受体的测定,推测龙胆碱对脑损伤保护作用的可能机制。方法 21日龄Wister大鼠100只,经筛选后随机分为空白对照组、模型组(FS组)、龙胆碱高剂量组(GT1组)、龙胆碱低剂量组(GT2),热水浴法诱导高热惊厥模型,采用茚三酮染色法,借助全自动生化分析仪,计算脑组织氨基酸含量:氨基酸含量(mg/g湿组织)=每20μl上清液中氨基酸的含量/20×上清液总量(μl)/脑组织湿重(g)。免疫组化法观察海马区GABAB、Glu受体蛋白表达变化。结果 GT各组大鼠脑组织中Glu含量高于模型组,GABA含量低于FS组,Asp、Gly与FS组无显著性差异。GT1、GT2与FS组相比,差异具有显著性(P<0.05);GABA蛋白表达,GT1、GT2组在海马CA1~CA3区GAB-AB蛋白表达明显高于FC组,但低于对照组,差异具有显著性;Glu各组间比较,差异均无显著性。结论龙胆碱对惊厥所致的脑损伤的保护作用机制与脑内神经递质GABA、Glu改变有关,GABA递质的增高与GABAB受体的相应增加是其作用的机制之一。Objective To study possible mechanisms of the protective effects on brain damage of gentianine.Methods 21-day old wistar rats were randomly divided into 4 groups:Normal control group(n=15),FS group(n=20),gentianine high-dose group(GT1 group,n=20),gentianine low-dose group(GT2,group.n=20).Automatic amino acid analyzer was used to observe the content changes of exactatory-inhibitory amino acid neurotransmitters(Glu,Asp,GABA,Gly),and protein expression of GABABR,Glu receptor in hippocampus were detected by immunohistochemistry.Results The contents of GABA in the GT1、GT2 group were those of glu were lower FS group,the difference was significant between GT1、GT2 groups and FS group but the contents of Asp,Gly had no significant difference from FS group.The reslut of GABABR showed that the expression of GABAB was higher than FS group but lower than NG group in the GT1.GT2 group,and the difference was significant,but the GluR had no significant difference.Conclusion The protective effect on th...更多e damage of brain of gentianine is related to the change of GABA and Glu contents in the rat brain,one of pharmacological mechanism on brain damage protection of gentianine is to increase the protein expression of GABABR and the content of GABA and decrease the contents of Glu in brain 还原
