蒿鳖养阴软坚方中大黄素含量的高效液相色谱法测定

摘　　要：目的建立测定蒿鳖养阴软坚方中大黄素含量的高效液相色谱(HPLC)方法。方法采用HPLC法,色谱柱HypersilODS2(250mm×4.6mm,5μm),流动相:甲醇-0.1%磷酸(75∶25);流速:1.0ml.min-1;检测波长:254nm。结果大黄素在2.64～26.4μg.ml-1范围内呈良好线性关系,r=0.9998,平均加样回收率为99.7%,RSD为2.7%(n=6)。结论该方法可将大黄素从复方中充分提取分离,专属性强,结果准确,可用于蒿鳖养阴软坚方的质量控制。Objective To establish an HPLC method for the determination of emodin in Hao Bie Yang Yin Ruan Jian Fang.Methods The emodin in Hao Bie Yang Yin Ruan Jian Fang was determined by HPLC.Hypersil ODS2 column（250 mm×4.6 mm）was used,0.1% phosphoric acid：water（75∶25）was used as the mobile phase.The flow rate was 1.0 ml·min-1,and the detection wavelength was set at 254 nm.Results The linear range of emodin was 2.64～26.4 μg·ml-1,and the correlation coefficient was 0.9998,the average recovery rate was 99.7%,and RSD=2.7%（n=6）.Conclusion The established method is accurate,sensitive and simple.It can be used for quality control of Hao Bie Yang Yin Ruan Jian Fang.
