眼针疗法对肠易激综合征模型大鼠结肠组织5-羟色胺4受体表达的影响

摘　　要：目的研究肠易激综合征(IBS)模型大鼠远端结肠组织5-羟色胺4受体(5-HT4R)的表达以及眼针干预的影响。方法实验动物分成正常对照组、IBS模型组、眼针组,采用束缚刺激与应激刺激相结合的方法建立IBS动物模型并进行眼针干预。采用Western-Blot法、RT-PCR法检测各组大鼠远端结肠组织中5-HT4R的蛋白和mRNA表达。结果与正常对照组比较,IBS模型组5-HT4R的蛋白和mRNA表达均下调;而与IBS模型组比较,眼针组5-HT4R表达明显上调。结论 IBS模型大鼠结肠组织5-HT4R水平下调,提示5-HT4R可能是IBS分子生物学基础之一,为IBS的治疗提供了分子靶点。眼针干预可有效改善IBS大鼠5-HT4R表达水平。Objective To investigate the expression of 5-HT4 recepters of distal colonic mucosa in the rat with irritable bowel syndrome （IBS） and effects of ocular acupuncture therapy （OAT）. Methods Animals were classified into blank control, IBS model and OAT groups. Rat model of IBS was established by method of stress and bound, and OAT was given. The mRNA and protein content of 5-HT4 recepters of distal colonic mucosa of each group was examined by Western-Blot and RT-PCR. Results The mRNA and protein expression of 5-HT4 recepters in IBS model decreased remarkably compared with those in control. The expression of 5-HT4 recepters in OAT increased remarkably compared with those in IBS model. Conclusions The expression of 5-HT4 recepters is significantly decreased in IBS model, suggesting that 5-HT4 receptors are involved in the pathogenesis of IBS, which may offer a molecular target for treatment. OAT can improve 5-HT4 recepters function effectively.
