摘 要:采用刃天青显色法检测3种抗菌药物(头孢哌酮钠、链霉素、青霉素)对铜绿假单胞菌的MIC和最佳作用时间,并比较与微量稀释法的相关性,探讨应用该法的可行性。结果表明:刃天青的试验浓度在75—150ug/mL为宜.细菌接种量1×10 5cfu/孔,肉眼即可判定MIC值结果,判定时间为8h。3种抗菌药物对铜绿假单胞菌的MIC和最佳作用时间分别为:5ug/mL,4~6h;20ug,/mL,6~8h;不敏感。刃天青显色法与稀释法MIC值检测结果一致。本试验结果说明,刃天青显色法具有简便、快速、灵敏度高的特点,能动态地反映细胞随时间变化的生长情况,能检测药物的最佳作用时间。A resazurin-based assay was employed to evaluate MIC and the optimal function time of anti- P. vaeruginosa drugs in the current work. The results demonstrated that under the optimal condition of 75 - 150 ug/mL of resazurin, 1 × 105 cfu/well of bacteria inoculation density, and 8 hours of incubation, the MIC could be judged by naked eye. The MIC for cefoperazone sodium, streptomycin, or penicillin were 5 ug/ml, 20 ug/ml or un-sensitive, respectively. The optimal function times for cefoperazone sodium and streptomycin were 4 -6 h, 6 -8 h, respectively. The results from the resazurin -based assay were consistent with that obtained using conventional dilution assay. Therefore, resazurin assay may dramatically detect the cell growth and evaluate MIC and the optimal function time of anti-bacteria drugs. This resazurin -assay has the considerable advantages to the other assays in both simplicity and convenience.
